A BlackBerry Enterprise Server (BES) is attached to a company's mail sever. It provides instant email delivery as well as syncing or contacts, memos, and calendar events between the desktop mail client and the device, without having to connect the device via USB. Enterprise Activation is the process which tells the BES which device is connected to which mailbox.
If you are not using an enterprise server, it is of no use to you and you should just hide the icon.
Enterprise activations is if you are connecting your BB to your company servers to get email. You would get a password from your employers IT dept to connect. This is mostly for business users. Not all companies have the BES.
