Question Unable to Connect With Desktop Manager

Hi. I have tried everything to connect to my desktop. When I click on connect the screen is pointing to COM3 but I have the Blackberry plugged into a USB port. I select AUTO detect with no luck. The phone is causing a noise in the PC but always shows disconnected. In OPTIONS I cannot find the Communication Settings option one pop-up wants me to change. What do I do to connect to the desktop manager?
Thanks all.

A connection to the device could not be established. Please ensure that the device is properly connected to the cradle or USB cable, and that the cradle or USB cable is connected securely to your computer. Also ensure that the device has completed any startup operations. Then run the Application Loader Wizard again.
