Open BlackBerry assistant, swipe down from the top of the screen, settings, voice response on/off.

Touch the bar that says Voice Response,
From there you can toggle on or off.

On your home screen, touch the magnifying glass at the bottom. Now you are in the BlackBerry assistant. Swipe down from the top, settings...

Reset your device by holding down the up and down volume buttons (or maybe the power button) for 30 seconds or so, then try again once it restarts.

Grab yourself an autoloader for your device, download it and then install it. At this point not too many other options available since you stated that the settings for Assistant just brings you to the standard settings screen.

I'll have to agree with MobileMadness here, it looks like you'll have to reload your device with an Autoloader or BlackBerry Link if you prefer. Looks like your existing installation is somehow damaged.
Make sure you back up everything important before the reload.

Well you are on the official release so, unless you have particular reason to change, I would just reload it again.
Personally I would just backup my device using BlackBerry Link, make a further backup of the most important stuff by manually copying it somewhere as well, then just reload the OS using BlackBerry Link. After it has reloaded, restore your data, but avoid restoring your settings if you can, just reconfigure your device manually.
Others here will suggest using an Autoloader instead of BlackBerry Link, personal preference (when loading the current official) .
